ABSTRACT

@#[摘 要] 酪蛋白激酶2(CK2)是一种高度保守的丝/苏氨酸蛋白激酶,它能够促进肿瘤细胞增殖、分化、侵袭和转移,并协助肿瘤细胞抗凋亡,亦有助于形成肿瘤免疫抑制微环境。近年来多项研究结果表明,CK2在乳腺癌细胞中呈过表达状态,是乳腺癌发生与发展过程中不可或缺的蛋白激酶。目前,CK2作为乳腺癌的新型治疗靶点,其在国内外的相关研究相对较少,但研发的各型CK2抑制剂已展现出抑制乳腺癌的巨大潜能。阐明CK2的结构和生物学功能及其在乳腺癌发展中的作用机制,以及CK2抑制剂的开发和临床应用现状,将有助于以CK2为靶点的乳腺癌靶向治疗。

ABSTRACT

HyperCKemia is a rare condition characterized by a persistent increase in serum creatine kinase (CK) levels or some isoenzymes. Usually, there are no clinical, electromyography or histological manifestations, which involves a challenge at the time of diagnosis. The patient in question showed no characteristic signs or symptoms, apart from fatigue and post-exercise myalgia. Assessment was performed by rheumatology and endocrinology, determination of total CK and MB fraction in blood, and electromyography and protein electrophoresis were requested as part of the approach. This case report is considered as novel, interesting, and useful for clinical practice as few similar ones were found in the scientific literature. The difficult etiological diagnosis of this entity, and the algorithm used to arrive at it, are all presented. It is concluded that in those patients with hyperCKemia of unknown etiology, this diagnosis should be kept in mind, and be confirmed by performing a CK electrophoresis.


La hiperCKemia es una condición poco frecuente caracterizada por un aumento persistente de los niveles de creatina quinasa (CK) sérica o de algunas isoenzimas, sin que suelan presentarse manifestaciones clínicas, electromiográficas o histológicas, lo cual implica un desafío a la hora del diagnóstico. El paciente cuyo caso se presenta aquí no mostró signos o síntomas característicos, únicamente fatiga y mialgias posteriores al ejercicio. Se llevó a cabo valoración por reumatología y endocrinología, determinación de CK total y fracción MB en sangre; además, se solicitó electromiografía y electroforesis de proteínas como parte del abordaje. Consideramos que este reporte de caso es novedoso, interesante y de utilidad para la práctica clínica pues se encuentran pocos similares en la literatura científica; adicionalmente, se pone en evidencia el difícil diagnóstico etiológico de esta entidad, así como el algoritmo utilizado para llegar a ella. Se concluye que este diagnóstico debe tenerse en mente en aquellos pacientes con hiperCKemia de etiología desconocida, y para confirmarlo es necesario hacer una electroforesis de CK.

ABSTRACT

A muscle strain develops when a muscle lengthens or is torn. This would be frequently caused by muscle exhaustion, abuse, or misuse. Exercise-Induced Muscle Damage (EIMD) is defined based on indicators appearing soon after initiating an exercise session and continues for up to 14 days after the end of the session. Muscle damage from trauma, inherited genetic illnesses, pathology or complete circumstances are highly common and cause significant socio-economic consequences. Athletes care about EIMD-related loss of muscle strength and discomfort since they can affect their performance significantly. This current review is intended to highlight the updated findings and its present preventive and management strategies. The review also discussed the pathophysiology behind EIMD in detail to highlight the understanding points regarding EIMD. The frequency of EIMD following exercise must are linked with strength training, and muscle strain as measured by an increased level of serum kinase (CK). the percentage of tissue micro-injuries. EIMD is quite frequent among athletes and also among the general population. This review has discussed the available diet and nutrition to prevent such EIMD and tackle, it if EIMD sets in. Finally, this current review has covered EIMD from all the phases and recommendations have been made for further study, especially in clinical trials.

RESUMEN Se analizaron los niveles séricos de creatina quinasa-MB (CK-MB) y lactato deshidrogenasa (LDH) en 10 perros diagnosticados con enfermedad valvular degenerativa y en seis perros clinicamente sanos, con el objetivo de evaluar si sus niveles séricos indican daño miocárdico. Las muestras de suero se analizaron mediante el método UV. Se utilizó la prueba de diferenciación de medias para determinar diferencias entre medias, y la prueba de correlación de Pearson para determinar si existe correlación entre los niveles séricos de ambas enzimas. Los valores de CK-MB y de LDH fueron significativamente diferentes entre los dos grupos de pacientes. Los niveles de CK-MB y LDH tuvieron correlación positiva, pero no significativa.


ABSTRACT Serum levels of creatine kinase-MB (CK-MB) and lactate dehydrogenase (LDH) were analyzed in 10 dogs diagnosed with degenerative valvular disease and in six clinically healthy dogs with the objective of evaluating whether their serum levels indicate myocardial damage. Serum samples were analyzed by UV method. The mean differentiation test was used to determine differences between means and the Pearson correlation test was performed to determine if there was a correlation between the serum levels of both enzymes. The CK-MB and LDH values were significantly different between the two groups of patients. The levels of CK-MB and LDH had a positive but not significant correlation.

ABSTRACT

Context: Incidence of periampullary carcinoma is low, approximately 0.5–2% of all gastrointestinal malignancies. Histologic subtyping has a prognostic bearing. The purpose of this study is to differentiate periampullary carcinomas based on immunohistochemistry (IHC) by using cytokeratin 7 (CK7), cytokeratin 20 (CK20), caudal type homeobox 2 (CDX2). Aims: To analyze the usefulness of IHC as single/panel of markers that included CK7, CK20, and CDX2. Settings and Design: This was a prospective study done from January 2017 to September 2018. Subjects and Methods: A total 50 pancreaticoduodenectomy specimens were evaluated and classified as intestinal (INT) and pancreaticobiliary (PB) types based on their morphological and immunohistochemical features, respectively. The morphologic subtypes, expression of IHC markers were correlated with different histologic parameters. Statistical Analysis: Chi-square test was used to study the association between different IHC markers with histologic parameters. Probability (P) values <0.05 were regarded as statistically significant. Results: The expression of CK7, CK20, CDX2 were studied in 50 cases to classify them as INT and pancreatobiliary subtypes. CK7 has high sensitivity (88.2%), CDX2 has high specificity (96.4%), CK20+/CDX2+ has both high sensitivity (94.2 percent) and specificity (89.2 percent) in differentiating INT from pancreatobiliary subtypes. The morphologic subtypes showed correlation with two variables (tumor grade, pathologic T stage). CK20 and CK20/CDX2 expression showed a positive correlation with tumor grade, pathologic T staging, and lymphovascular invasion. Conclusions: In conclusion, morphological classification can significantly discriminate histologic types, IHC plays a moderate role. However, the combined expression of CK20 and CDX2 is helpful in subtyping.

ABSTRACT

The dysregulation of transcription factors is widely associated with tumorigenesis. As the most well-defined transcription factor in multiple types of cancer, c-Myc can transform cells by transactivating various downstream genes. Given that there is no effective way to directly inhibit c-Myc, c-Myc targeting strategies hold great potential for cancer therapy. In this study, we found that WSB1, which has a highly positive correlation with c-Myc in 10 cancer cell lines and clinical samples, is a direct target gene of c-Myc, and can positively regulate c-Myc expression, which forms a feedforward circuit promoting cancer development. RNA sequencing results from Bel-7402 cells confirmed that WSB1 promoted c-Myc expression through the β-catenin pathway. Mechanistically, WSB1 affected β-catenin destruction complex-PPP2CA assembly and E3 ubiquitin ligase adaptor β-TRCP recruitment, which inhibited the ubiquitination of β-catenin and transactivated c-Myc. Of interest, the effect of WSB1 on c-Myc was independent of its E3 ligase activity. Moreover, overexpressing WSB1 in the Bel-7402 xenograft model could further strengthen the tumor-driven effect of c-Myc overexpression. Thus, our findings revealed a novel mechanism involved in tumorigenesis in which the WSB1/c-Myc feedforward circuit played an essential role, highlighting a potential c-Myc intervention strategy in cancer treatment.

Objective To investigate the expression of CK2α' in hepatocellular carcinoma (HCC) tissues and its prognosis value. Methods Tissue microarray was made from tumor and adjacent normal tissues of 83 patients with HCC. Immunohistochemical was used to stain the microarray for semi-quantitative analysis, and the expression of CK2α' in HCC tissues and adjacent normal tissues was compared. The associations between CK2α' expression and clinicopathologic features of HCC patients were analyzed with χ2 text. The prognostic value of CK2α' was estimated using Kaplan-Meier approach. Results The average positive expression rate of CK2α' was 47.8% in HCC and 65.4% in adjacent normal tissues. The positive expression rate of CK2α' in HCC tissues was lower than that in adjacent normal tissues (P < 0.01). The expression level of CK2α' in the cancer tissues of 74.7% cases was significantly lower than that in the adjacent tissues. HCC patients with low CK2α' expression had higher risk of recurrence and shorter overall survival time (P < 0.05). The expression of CK2α' was significantly correlated with CK19 (P < 0.05). Conclusion CK2α' is down-regulated in HCC tissues and correlated with the prognosis of patients. It could be a potential prognostic marker for HCC.

Objective:To investigate CK19 and COX-2 expression in papillary thyroid carcinoma (PTC) and their relationship with clinicopathologic parameters.Methods:Retrospective study of 120 consecutive patients with PTC who underwent resection from May. 2017 to Dec. 2020 was conducted. The expression of CK19 and COX-2 in 120 pieces of primary PTC tissue and 30 pieces of adjacent carcinoma tissue were detected by EliVision TM plus two-step immunohistochemical method. The relationship between the expression of CK19 and COX-2 and the clinicopathologic parameters including patient age, TI-RADS classification, TNM staging, carcinomatous infiltration, lymph node metastasis was studied. The analysis was performed using SPSS 22.0 software. The numerical data were presented as numbers and percentages, and chi-test ( χ2 test) and Pearson’s correlation were used to analyze the difference and association between two groups. P<0.05 was considered statistically significant. Results:The positive immunostaining of CK19 and COX-2 were mainly localized in the cytoplasm. The positive rates of CK19 and COX-2 were 87.50% (105/120) and 72.50% (87/120) in cancer tissues and 10.00% (3/30) and 3.33% (1/30) in paracancerous tissues, respectively, with statistically significant differences (both P<0.05 by χ2 test) . In the groups with patients aged <45 years versus ≥45 years, the CK19 positive rates were 88.16% (67/76) and 86.36% (38/44) , and the COX-2 positive rates were 69.74% (53/76) and 77.27% (34/44) , respectively. In the groups of TI-RADS (grade 4 and 5) versus grade 6, the CK19 positive rates were 85.26% (81/95) and 96.00% (24/25) , and the COX-2 positive rates were 69.47% (66/95) and 84.00% (21/25) , respectively, with no statistically significant differences between the two groups (both P>0.05) . In the groups of TNM (stage T1 and T2) versus stage T3, the CK19 positive rates were 82.28% (65/79) and 97.56% (40/41) , and the COX-2 positive rates were 65.82% (52/79) and 85.36% (35/41) , respectively. In the groups with versus without cancer infiltration, the CK19 positive rates were 94.44% (68/72) and 77.08% (37/48) , and the COX-2 positive rates were 83.33% (60/72) and 56.25% (27/48) , respectively. In the groups with versus without lymph node metastasis, the CK19 positive rates were 95.59% (65/68) and 76.92% (40/52) , and the COX-2 positive rates were 83.82% (57/68) and 57.69% (30/52) , respectively, and the differences between the above groups were statistically significant (all P<0.05 by χ2 test) . The co-positive and co-negative expression rates of CK19 and COX-2 in 120 patients were 70.83% (85/120) and 10.83% (13/120) , respectively, with a positive correlation ( r=0.45, P<0.05 by Pearson’s correlation) . Conclusions:The positive rates of CK19 and COX-2 expressions are not related to patient’s age or TI-RADS classification, but closely related to TNM stage, cancer invasion and lymph node metastasis. The up-regulation of both CK19 and COX-2 expressions predicts higher tumor stage, more aggressive invasion and more prone to lymph node metastasis.

ABSTRACT

Protein kinase CK2 is a common, evolutionarily conserved and ubiquitous protein kinase. In recent years, increasing evidences have shown that CK2 has a variety of phosphorylated protein substrates, which play important roles in growth, development and various diseases. Therefore, CK2 may participate in these physiological processes by regulating the phosphorylation of these substrates. This article briefly reviewed the structural characteristics of protein kinase CK2 and its physiological functions in growth, development, immunity, formation of tumor and other diseases, in order to provide knowledge basis for further research on the regulatory mechanism of CK2 and potential applications of its inhibitors.

ABSTRACT

Hepatocellular carcinoma (HCC) has been known as the second common leading cancer worldwide, as it responds poorly to both chemotherapy and medication. Triptolide (TP), a diterpenoid triepoxide, is a promising treatment agent for its effective anticancer effect on multiple cancers including HCC. However, its clinical application has been limited owing to its severe systemic toxicities, low solubility, and fast elimination in the body. Therefore, to overcome the above obstacles, photo-activatable liposomes (LP) integrated with both photosensitizer Ce6 and chemotherapeutic drug TP (TP/Ce6-LP) was designed in the pursuit of controlled drug release and synergetic photodynamic therapy in HCC therapy. The TP encapsulated in liposomes accumulated to the tumor site due to the enhanced permeability and retention (EPR) effect. Under laser irradiation, the photosensitizer Ce6 generated reactive oxygen species (ROS) and further oxidized the unsaturated phospholipids. In this way, the liposomes were destroyed to release TP. TP/Ce6-LP with NIR laser irradiation (TP/Ce6-LP+L) showed the best anti-tumor effect both

ABSTRACT

RAS, a member of the small GTPase family, functions as a binary switch by shifting between inactive GDP-loaded and active GTP-loaded state. RAS gain-of-function mutations are one of the leading causes in human oncogenesis, accounting for ∼19% of the global cancer burden. As a well-recognized target in malignancy, RAS has been intensively studied in the past decades. Despite the sustained efforts, many failures occurred in the earlier exploration and resulted in an 'undruggable' feature of RAS proteins. Phosphorylation at several residues has been recently determined as regulators for wild-type and mutated RAS proteins. Therefore, the development of RAS inhibitors directly targeting the RAS mutants or towards upstream regulatory kinases supplies a novel direction for tackling the anti-RAS difficulties. A better understanding of RAS phosphorylation can contribute to future therapeutic strategies. In this review, we comprehensively summarized the current advances in RAS phosphorylation and provided mechanistic insights into the signaling transduction of associated pathways. Importantly, the preclinical and clinical success in developing anti-RAS drugs targeting the upstream kinases and potential directions of harnessing allostery to target RAS phosphorylation sites were also discussed.

@#[Abstract] Objective: To investigate the effect of cytokeratin 13 (CK13) on radio-sensitivity of human nasopharyngeal carcinoma HNE1 cell line and its mechanism. Methods: HNE1 cells were divided into control group, anti-CK13#a group (CK13 knockdown), anti-CK13#b group (CK13 knockdown), control+sirolimus group (100 nmol/L sirolimus treatment for 1 h), and anti-CK13#a + sirolimus group (100 nmol/L sirolimus treatment for 1 h). After irradiation treatment (200 cGy/min irradiation for 5 min), cell proliferation in each group was measured by CCK-8 assay. Cell apoptosis rate in each group was determined by Flow cytometry. Expression of PI3K/AKT/mTOR signaling pathway related PTEN gene was detected by qPCR, and WB was used to detect the expressions of PI3K/AKT/mTOR signaling pathway related proteins. Results: In the case of radiotherapy, as compared with the control group, the proliferation of HNE1 cells after CK13 knockdown was significantly enhanced (P<0.01) while the apoptosis rate was significantly reduced (P<0.01), the contents of caspase-3 and γH2AX as well as the protein lever of PTEN in cells were significantly decreased, while the expressions of p-AKT and p-S6K were significantly increased (all P<0.01). Interestingly, additional treatment with sirolimus (PI3K/AKT/mTOR signaling pathway inhibitor) could rescue the accelerated cell proliferation and decreased cell apoptosis caused by CK13 knockdown (all P<0.05). Conclusion: CK13 knockdown can enhance the activity of PI3K/AKT/mTOR signaling pathway by down-regulating PTEN, and ultimately reduce the radio-sensitivity of nasopharyngeal carcinoma HNE1 cells.

Resumen El accidente ofídico representa un importante problema de salud pública en las zonas tropicales y subtropicales del mundo. La cascabel común (Crotalus durissus cumanensis) es la serpiente del género Crotalus más abundante en Venezuela. El objetivo fue determinar los cambios séricos en la alanino aminotransferasa (ALT), aspartato aminotransferasa (AST), lactato deshidrogenasa (LDH) y creatina fosfocinasa (CK) en ratas, inducidos por el veneno de Crotalus durissus cumanensis, así como el establecimiento de los efectos histopatológicos. Se prepararon cinco grupos experimentales con tres ratas cada uno, reservando un grupo control placebo (G1), el cual fue tratado por ruta IP con 100 µL de solución salina fisiológica estéril (SSFe). Al resto de los animales se les administraron 50 µg de veneno nativo por la misma vía. Estos fueron sacrificados a las 1 (G2), 3 (G3), 6 (G4) y 9 (G5) horas posinoculación, con tomas de muestras séricas para determinaciones de las referidas enzimas y de diferentes órganos para estudios histopatológicos (corazón, músculo esquelético, hígado y riñón). Se observó un incremento en la ALT (p < 0,005) a partir de la primera hora posinyección, con un pico a la novena hora de 147 ± 8 UI/L; para la AST (p < 0,005) a partir de la primera hora posinyección con concentración pico a la sexta hora (293 ± 8 UI/L). De la LDH (p < 0,005) se registró un pico máximo de 2700 ± 8 UI/L a la novena hora posinyección. La CK (p < 0,005) mostró un pico máximo de concentración a la sexta hora (1489,66 ± 8,5 UI/L). Los resultados histopatológicos en tejido cardíaco mostraron hiperemia, congestión y necrosis de Zenker; en músculo esquelético, hiperemia, infiltrado inflamatorio, necrosis de Zenker; en hígado, extravasación de glóbulos rojos, congestión de sinusoides, telangiectasia e infiltrado inflamatorio en vena centrolobulillar. El riñón mostró hemorragia en túbulos contorneados, infiltrado inflamatorio, colapso de los túbulos hasta la pérdida de la arquitectura del órgano. Los efectos observados fueron tiempo-dependientes. Los resultados pueden indicar un posible efecto miotóxico y hepatotóxico.


Abstract The ophidian accident represents a major public health problem in the tropical and subtropical areas of the world. The common rattlesnake (Crotalus durissus cumanensis) is the most abundant snake of the genus Crotalus in Venezuela. The objective was to determine the serum changes in alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH) and creatine phosphokinase (CK) in rats, induced by the poison of Crotalus durissus cumanensis, as well as the establishment of histopathological effects. Five experimental groups were prepared with 3 rats each, reserving a placebo control group (G1), which was treated by IP route with 100 µL of sterile Physiological Saline Solution (SSFe), the rest of the animals were administered 50 µg of venom native by the same route, which were sacrificed at 1 (G2), 3 (G3), 6 (G4) and 9 (G5) hours post-inoculation, with serum samples taken for determinations of said enzymes, as well as different orgns for histopathological studies (heart, skeletal muscle, liver and kidney). An increase in ALT (p <0.005) was observed from the first hour post injection, with a peak at the ninth hour of 147 ± 8 IU / L; for AST (p <0.005) from the first hour post-injection with peak concentration at the sixth hour (293 ± 8 IU / L); with respect to LDH (p <0.005), a maximum peak of 2700 ± 8 IU / L was recorded at the ninth hour post-injection. CK (p <0.005) registered a maximum concentration peak at the sixth hour (1489.66 ± 8.5 IU / L). Histopathological results in cardiac tissue showed Zenker hyperemia, congestion, and necrosis; in skeletal muscle hyperemia, inflammatory infiltrate, Zenker's necrosis; in liver extravasation of red blood cells, sinus congestion, telangiectasia and inflammatory infiltrate in the centrilobular vein; the kidney showed hemorrhage in contoured tubules, inflammatory infiltrate, collapse of the tubules until loss of the organ architecture. The observed effects were time-dependent. The results could indicate a possible myotoxic and hepatotoxic effect.

The incidence of polycystic ovary syndrome is found to be very common in patients attending gynaecology department. The incidence is growing rapidly among young adults. PCOS leads to multiple complications. A study of 120 women with PCOS was done in Kerala in India in order to analyse the prevalence, clinical features and impact on life.METHODSCase records of women who were diagnosed as Polycystic Ovarian Disease who attended the gynaecology OPD in Karuna Medical College Hospital in Kerala were randomly selected. Women included in the study were in the age group of 14 – 30 years. The sample size selected was 120. The study was carried out for a period of 12 months in our hospital with the help of the records available. The diagnosis was made as per Rotterdam’s criteria.RESULTS75 women were unmarried and 45 married. 2 of the women had subclinical hyperandrogenism. 70 women have DHEA increased. FSH: LH ratio > 2:1 seen in 84 women. Testosterone was normal. Anti-Mullerian Hormone was raised in 84 women, indicating hyperandrogenism. Serum Prolactin was normal in most women except in 2 women.CONCLUSIONSPCOD leads to many complications like diabetes, Metabolic X syndrome, cardiovascular complications, breast cancer, endometrial cancer and depression. Kerala is considered as the diabetic capital in India. There are several risk factors like, family history, lack of exercise, stress, and high carbohydrate diet. A screening programme for PCOD is essential to prevent the sequelae.

Adult body harbors powerful reservoir of stem cells that maintains homeostasis by tissue regeneration and in response to disease and injury. Hair follicle is a dynamic mini organ supporting important biological functions of the body in maintaining homeostasis and skin tissue self-renewal. This study was carried out with the objective of finding the adult stem cells in canine hair follicular tissue. To conduct this study, adult canine skin samples (n=12) irrespective of breed and sex were collected. To characterize the hair follicle stem cells, paraffin sections of canine hair follicles were immunostained with positive hair follicle stem cell markers like Anti- cytokeratin 15 (CK15) and Anti-cytokeratin 19 (CK19) and FITC conjugated and HRP conjugated secondary antibodies were used. Immunoreactivities for CK15 and CK19 were observed in the bulge/isthmus region of hair follicles in between the infundibulum and suprabulbar regions and occupied most part of the peripheral layer of outer root sheath cell. Immunophenotyping of canine Hair Follicle Stem Cells (cHFSCs) in the bulge region of hair follicle helps in confirmation of in vitro culture of cHFSCs from the bulge region which will be further used for translational research

Objective@#To detect the expression and the differential significance of CK5/6, DSG3, P40, TTF-1, CK7, NapsinA in small biopsy specimens of non-small cell lung cancer (NSCLC), squamous cell carcinoma (SCC) and adenocarcinoma (AC).@*Methods@#Immunohistochemical SP method was used to detect the expressions of CK5/6, DSG3, P40, TTF-1, CK7 and NapsinA in 120 small biopsy specimens of NSCLC hospitalized in the Central People's Hospital of Tengzhou from January 2016 to December 2017, and the results were analyzed combined with the clinical characteristics of NSCLC.@*Results@#The positive expression rates of CK5/6, DSG3 and P40 in lung SCC were 100.0%(56/56), 89.3%(50/56) and 96.4%(54/56), respectively, with specificity of 90.6%, 100.0% and 100.0%, respectively.The positive expression rates of NapsinA, TTF-1 and CK7 in lung AC were 81.3%(52/64), 90.6%(58/64) and 93.8%(60/64), respectively, with specificity of 100.0%, 92.9% and 96.4%, respectively.The positive expression rates of CK5/6, DSG3, P40 in SCC had statistically significant differences compared with those in AC (all P<0.05), and the expression of TTF-1, CK7 and NapsinA in AC had statistically significant differences compared with those in SCC (all P<0.05).@*Conclusion@#CK5/6, DSG3, P40, TTF-1, CK7 and NapsinA are of great significance in the differential diagnosis of SCC and AC in small biopsy specimens of NSCLC.

ABSTRACT

@#[Abstract] Objective: :To investigate the effect of miRNA-325-3p and its target gene cytokeratin 13 (CK13) on the radio-sensitivity of nasopharyngeal carcinoma cell line CNE1. Methods: :The potential target gene of miRNA-325-3p was predicted by three databases: miRBase, Targetscan and microcosm, and verified by Double luciferase activity assay. QPCR was used to detect the expression levels of miRNA-325-3p and its target gene in nasopharyngeal carcinoma cell line CNE1 under different radiation doses; To verify the changes in radio-sensitivity of nasopharyngeal carcinoma cells, colony formation assay, Flow cytometery and MTT were used to observe the clone formation, apoptosis and cell viability of CNE1 cells after overexpression of miRNA-325-3p and knockdown of CK13 under different radiation doses, respectively. Results: :CK13 was confirmed as a potential target gene of miRNA-325-3p. After radiotherapy, the expression level of miRNA-325-3p in CNE1 cell was significantly increased, while the expression level of CK13 was decreased (all P<0.05); up-regulation of miRNA-325-3p expression and silence of CK13 gene increased cell survival rate (upregulation of miRNA325-3p: [60.14±3.55]% vs [19.23±3.42]%, t=14.37, P<0.01; silence of CK13: [76.15±5.13]% vs [28.53±3.68]%, t=13.06, P<0.01) and colony formation rate, and decreased apoptosis rate (upregulation of miRNA-325-3p: [27.95±2.67]% vs [51.68±3.47]%, t=9.39, P<0.01; silence of CK13: [20.31±2.62]% vs [38.14±3.83]%, t=6.66, P<0.01). Conclusion: :miRNA-325-3p can reduce the sensitivity of nasopharyngeal carcinoma cell line CNE1 to radiotherapy by down-regulating the target gene CK13.

Background: India has shown a sharp rise in the number of suicidal poisoning in the past few years and organo-phosphorus compounds are the most commonly implicated. This is a prospective study done to assess if troponins can be used as a prognostic marker in patients with organophosphate poisoning.Methods: Author conducted a prospective study on 50 patients to measure the level of troponin T and CK-MB on admission and Day 3 in patients with OP poisoning. This was then correlated to the severity of poisoning and also studied for its association with outcomes like respiratory failure or death. The primary endpoints were death and respiratory failure while secondary endpoint was the length of ICU stay.Results: With 0.1 ng/ml taken as the cut off for Troponin- T, and 40 U/L for CK-MB, positive cardiac enzymes were seen only in 10% of the patients, but all the patients in this group developed res-piratory failure. The mean Troponin T and CK-MB levels (0.1142±0.06 ng/ml and 39.14±8.23 U/L) were higher for patients who died when compared to the survivors. A similar picture was seen in patients with respiratory failure (Troponin T 0.0895±0.01 ng/ml and 34.65±11.66 U/L) as compared to those without respiratory failure.Conclusions: The level of cardiac enzymes correlated well with the severity of poisoning, days of ICU stay and outcome, thus suggesting its use as a prognostic indicator of organophosphorus poisoning.
